Реферат: eng: Air pollution by particulate matter (PM) has been associated with cardiopulmonary morbidity and mortality in many recent epidemiological studies. it has been shown that transition metal compounds, well-known toxic components of PM, are able to induce hypothermia following whole-body inhalation exposure. Low temperature appears to protect tissue against toxic effects of PM metal compounds in vivo and in vitro. To study the role of soluble and insoluble irritants in the induction of the hypothermic response, we analyzed the decrease in mouse body temperature (Δt body) after intranasal administration of PtO nanoparticles or a K 2[PtCl 4] solution. Between-strain differences in Δt body after intranasal administration of the irritants were evaluated using 6 inbred (BALB/cJ, C57BL/6J, AKR/OlaHsd, DBA/2JRccHsd, C3H/HeNHsd, and SJL/J) and 2 outbred mouse strains (SCiD and CD1). BALB/cJ and SCiD mice showed the most pronounced effect of intranasal administration of the xenobiotic on t body. Thus, t body was significantly lower after nasal administration the PtO nanoparticles than after administration of the K 2[PtCl 4] solution. To study the mechanism of this decrease, we compared the respective values for Δt body following intranasal, intravenous and per oral administration of PtO nanoparticles in Balb/c mice. Neither intravenous nor per oral administration had any effect on mouse body temperature. This fact together with data on the dynamics of the decrease in mouse body temperature following after intranasal administration of PtO nanoparticles (max Δt body ~ 80-100 min) allowed us to assume that this process is under nervous regulation. The correlation found between our data and some well-known phenotypic characteristics (phenome.jax.org) of the mouse strains used confirms this hypothesis.
rus: Одними из компонентов пыли с доказанными токсическими эффектами на дыхательную и кардиоваскулярную системы являются различные соединения тяжелых металлов. Известно, что при воздействии тяжелых металлов на дыхательную систему происходит понижение температуры тела животного, которое считается адаптивной реакцией, уменьшающей интенсивность проникновения токсиканта. Для изучения вклада растворимых и нерастворимых форм ксенобиотиков в развитие гипотермического ответа в данной работе проводили сравнение изменения температуры тела мышей в ответ на интраназальное введение коллоидного раствора наночастиц оксида платины (PtO-НЧ) и истинного раствора соли платины (K 2[PtCl 4]). Значение генетической изменчивости было исследовано на 6 инбредных (BALB/cJ, C57BL/6J, AKR/OlaHsd, DBA/2JRccHsd, C3H/HeNHsd, SJL/J) и 2 аутбредных линиях (SCiD и CD1) мышей. Наиболее выраженное снижение температуры в ответ на интраназальное введение ирританата продемонстрировали мыши линий BALB/cJ и SCID, при этом амплитуда падения температуры тела (Δt тела ) в ответ на введение PtO-НЧ была больше, чем на K 2[PtCl 4]. Механизм снижения температуры тела при интраназальном введении ксенобиотика определяли путем сравнения Δt тела для интраназального, внутривенного и перорального введения PtO-НЧ. Ни внутривенное, ни пероральное введение наночастиц не вызывало значимого изменения Δt тела животного, что вкупе с данными по динамике снижения температуры (max Δt тела ~ 80-100 мин) позволяет предположить опосредованный нервной системой механизм регуляции изменения Δt тела, который может реализоваться за счет активации афферентных волокон тройничного нерва. Корреляции данных по max Δt тела в ответ на интраназальное введение PtO-НЧ и ряда известных фенотических характеристик (phenome.jax.org) использованных нами линий подтверждают это предположение.
